Vitex negundo L. is a woody aromatic shrub distributed throughout India. All parts of the plant possess a multitude of secondary metabolites and finds use for treatment of a wide spectrum of health disorders in traditional and folk medicine. The plant has anti-inflammatory, anticonvulsant, antioxidant properties and are used for the treatment of various diseases such as asthma, bronchitis, cold, cough etc. In vitro studies and phytochemical analysis will be helpful for sustainable utilization of V. negundo. This work reports in vitro propagation of the plant along with evaluation of phytochemical constituents responsible for its unique properties and antimicrobial activity. In vitro studies were carried out in MS medium with various combinations of auxin and cytokinin. Various explants were tried for inducing callus, root and multiple shoots. Phytochemical screening of secondary metabolites in V.negundo was carried out along with evaluation of antimicrobial and antioxidant potential. This study presented a detailed knowledge about phytochemical constituents, antimicrobial & antioxidant properties along with a protocol for rapid in vitro propagation of Vitex negundo.

4. Role of growth hormones in rooting of Vitex negundo L: Studies on vegetative propagation of Chinese chaste tree (Vitex negundo L) by stem cuttings

A study on the vegetative propagation of Chinese chaste tree (Vitex negundo L.) was carried out at the Department of Horticulture, University of Agricultural Sciences, Gandhi Krishi Vignana Kendra, Bengaluru during August, 2010 to December, 2010. In the present experiment, the effect of growth regulators viz., IBA and NAA singly and in combinations on rooting of hardwood cuttings was studied. The treatments comprised of control, IBA at 2000 and 3000 ppm, NAA at 2000 and 3000 ppm, IBA+NAA 2000 ppm each and IBA+NAA 3000 ppm each and keradix. The experiment was conducted in a four replicated Factorial Completely Randomized Design. Single, double and triple node hardwood cuttings were treated with the growth regulator solutions. Observations were recorded on various shoot and root parameters and the survival percentage of rooted cuttings was also studied. The results of this investigation clearly brought out that, triple node hardwood cuttings should be treated with 3000 ppm IBA to obtain maximum rooting success and also for obtaining maximum survival of rooted cuttings in secondary nursery.

Vitex L. (Lamiaceae, Viticoideae) is taxonomically very complex comprising c. 250 pan-tropical, subtropical and a few temperate species. The most extensive taxonomic revision of the genus was by Pieper for African and Malagasy taxa and goes back to 1928. The need for a modern revision of the whole genus became critical with its recent transference from the Verbenaceae to the Lamiaceae (subfamily Viticoideae). Vitex is a large genus (3.6% of the Lamiaceae s.l.) and its taxonomic position is not of minor taxonomic/evolutionary importance. Vitex is a very important genus in traditional medicine throughout the tropics and especially in Africa. The scientific validation of its traditional medicinal uses requires a solid taxonomy. This book, therefore, provides the first molecular phylogeny reconstruction of the genus and provides explanation about the biogeographical patterns and dating of the molecular phylogeny. Also, an alpha-taxonomic revision of the grex Pilosae is provided based on morphological, molecular and chemical characters. The traditional uses in African medicine of some species were also validated by proving the bioactivities of the essential oils.

Vitex leucoxylon (Verbenaceae) is a deciduous (large and lofty) tree rarely found in tropical forests.V. leucoxylon(Kattu Nochi) is one of the underutilized medicinal species with great pharmacological potential. Its restricted natural distribution has limited its continued usage in indigenous systems of India. Phytochemical screening of all extracts of Vitex leucoxylon revealed the presence of steroids, terpenoids, tannins, saponins, flavonoids, alkaloids, cardiac glycosides, gums and mucilage. The essential oil content in leaves, flowers and fruits have been investigated by hydrodistillation method which confirmed the seasonal variation in leaves. In vitro antimicrobial evaluation confirms that the plant contains biologically active compounds which act against some pathogenic bacteria and fungi with some measurable antimicrobial response.

The results indicates the presence of active ingredients in the crude extracts.The ethyl acetate and ethanol extracts of fruit of Vitex negundo and bulb of Allium sativum could be used for further isolation and purification of novel lead molecules.This book, therefore provides further information regarding the bio-activity of these two plants and the results should be useful to professionals in drug discovery,or anyone else who may be interested to do research in medicinal plants.
